Genetic Maps of Stem Rust Resistance Gene Sr35 in Diploid and Hexaploid Wheat

چکیده
Puccinia graminis f. sp. tritici is the causal agent of stem rust of wheat. A new race designated TTKSK (also known as Ug99) and its variants (TTKST and TTTSK) are virulent to most of the stem rust resistance genes currently deployed in wheat cultivars worldwide. Therefore, identi! cation, mapping, and deployment of effective resistance genes are critical components of global efforts to mitigate this threat. Multipathotype seedling tests demonstrated that resistance gene Sr35 is effective against the three TTKS variants and another broadly virulent race from Yemen, TRTTF. Two genetic maps of Sr35 are presented in diploid (Triticum monococcum) and two in hexaploid wheat (T. aestivum). The Sr35 resistance to TRTTF and RKQQC races was mapped in diploid wheat within a 2.2 to 3.1 cM interval on the long arm of chromosome 3Am between markers XBF483299 and XCJ656351. This interval corresponds to a 174-kb region in Brachypodium that includes 16 annotated genes. The Sr35 map location was con! rmed in two backcross-derived hexaploid populations segregating for Sr35. Recombination between diploid and hexaploid chromosomes was 10-fold lower than between homologous chromosomes, but was suf! cient to reduce the introgressed diploid segment. These maps provide markers closely linked to Sr35 that will be useful to accelerate its deployment and pyramiding with other stem rust resistance genes. Genetics of resistance to race TTKSK of Puccinia graminis f. sp. tritici in Triticum monococcum.
Race TTKSK (or Ug99) of Puccinia graminis f. sp. tritici possesses virulence to several stem rust resistance genes commonly present in wheat cultivars grown worldwide. New variants detected in the race TTKSK lineage further broadened the virulence spectrum. The identification of sources of genetic resistance to race TTKSK and its relatives is necessary to enable the development and deployment o...

Wheat stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), is a devastating disease that can cause severe yield losses. A previously uncharacterized Pgt race, designated Ug99, has overcome most of the widely used resistance genes and is threatening major wheat production areas. Here, we demonstrate that the Sr35 gene from Triticum monococcum is a coiled-coil, nucleotide-binding, leucine...

Wheat stem rust caused by Puccinia graminis f. sp. tritici (Pgt) is one of the most important diseases of wheat, worldwide. New sources of resistance to stem rust could be found in synthetic hexaploid wheat (SHW) genotypes due to their high genetic diversity. In order to recognize the genetic structure of the population of the stem rust, following race analysis of 12 Pgt isolates, resistance of...

The hexaploid wheat (Triticum aestivum) adult plant resistance gene, Lr34/Yr18/Sr57/Pm38/Ltn1, provides broad-spectrum resistance to wheat leaf rust (Lr34), stripe rust (Yr18), stem rust (Sr57) and powdery mildew (Pm38) pathogens, and has remained effective in wheat crops for many decades. The partial resistance provided by this gene is only apparent in adult plants and not effective in field-g...

Stem or black rust of wheat, caused by Puccinia graminis Pers. f. sp. tritici Eriks. and Henn (Pgt), was the most feared and devastating disease of wheat at one time worldwide. The fear from stem rust was understandable because an apparently healthy looking crop about 3 weeks prior to harvest could reduce to a black tangle of broken stems and shriveled grain by harvest. The disease has been co...
